Relevance of the rat as a model for PSLT inhalation toxicology

  • Is the rat lung biological response to PSLTs (e.g., inflammation, hyperplasia, fibrosis, tumors) unique from other rodent species or humans?
  • Is the rat a relevant model of human lung cancer hazard and risk for inhaled PSLTs under conditions of lung particle overload?
  • If adverse lung effects (e.g., fibrosis, tumors) are observed in a PSLT study with rats only under conditions of lung particle overload how should the results be extrapolated to lower, non-overloading conditions?What findings support and what findings detract from the rat as relevant model to evaluate human health effects of inhaled PSLTs?

  • What is the most appropriate animal species for assessing human health hazards and risks from inhalation of PSLTs?What is the value of (negative) epidemiological outcomes of particles such as PSLT for current hazard classification?
